Cells were incubated for 48 h with 10 μM Nolvadex (Sigma-Aldrich, St. Louis, MO, USA) in the presence of varying concentrations of the drug in DMEM. Following incubation for 48 h, cells were stained with DAPI for histological analysis. The cells were visualized by confocal laser scanning microscopy (COSMAD, Requens, Berlin, Germany) at an excitation wavelength of 486 nm, which was used to evaluate the expression of the gene. The cells were examined by flow cytometry, with the cells stained for apoptosis. The cells were stained with the Annexin V-FITC apoptosis kit (Milane Biotech, San Jose, CA, USA) and propidium iodide (PI; Abcam, Cambridge, UK).

Results:A higher percentage of cells expressing the apoptosis-inducing marker, cleaved caspase-3 was observed in Ovariin-7-treated cells. This was associated with a decrease in the percentage of cells expressing the cleaved caspase-3 marker, cleaved-caspase-2, and an increase in the percentage of cells expressing the cleaved-caspase-3 marker, cleaved-caspase-3-AS. However, the apoptotic rate of Ovariin-7-treated cells was lower than that of control cells. Moreover, a higher percentage of apoptotic cells in Ovariin-7-treated cells than in control cells was observed in the presence of 10-mM Nolvadex (p < 0.05,p< 0.01,< 0.01). These results indicated that the treatment with nolvadex resulted in an increase in the number of apoptotic cells and an increase in apoptosis, indicating an increase in apoptosis.

Conclusion:Nolvadex was shown to increase the number of apoptotic cells and decrease the apoptotic rate of the Ovariin-7-treated cells. The increase in the apoptotic rate of Ovariin-7-treated cells was associated with an increase in the number of apoptotic cells. These results indicated that nolvadex reduced the number of apoptotic cells and an increase in apoptosis, suggesting that the mechanism of action of nolvadex is different from that of estrogen.
